The purpose of this study was to identify the changes in protein composition and sugar content of cotyledon during cold treatment of radish seedlings. After 3day old radish seedlings were incubated for various time at 6 ℃ or 10 ℃, protein extracts of cotyledon were analysed by SDS - PAGE. The protein composition of cold treated cotyledon was different from that of normal cotyledon. But,the protein composition was not different between 6 ℃ or 10 ℃ treated cotyledon. During cold treatment, 41, 51, and 60 kD proteins were significantly accumulated and persisted. In addition to these, smaller proteins whose molecular weight were 22, 26 and 34 kD increased transiently. On the other hand, 15.5, 16.5, 20, 23, and 30 kD proteins were decreased transiently during cold treatment. It has been suggested that low temperature, ABA and desiccation may have common effects on the protein synthesis. To test this hypothesis we studied the effect of ABA or desiccation on the protein composition of cotyledon of radish seedlings. We observed that the protein composition resulting from treatment of ABA and desiccation was similar to that resulting from cold treatment. To elucidate the mechanism through which the protein composition changed, we investigated the effect of actinomycin D and cycloheximide on the protein composition of the cotyledon of radish seedlings during cold treatment. Major changes appered more rapidly with cycloheximide and actinomycin D. We also studied the effect of cold treatment on the content of sucrose and reducing sugar of cotyledon. The content of sucrose increased within 1 day after 6 ℃ treatment. Then, it decreased for 2 - 3 days after cold treatment. It increased slowly there after.
