Studies on the new antibiotics-synthesis-suppression-mutant in streptomyces coelicolor

To define new genetic elements that regulate antibiotic synthesis, we screened for mutations that visibly blocked synthesis of the two pigmented antibiotics and found mutant strains in which all four antibiotics were blocked. We found mutant strains which we had named abs8752 and abs95. Both of them seems different from absA and absB mutants, which were found before. The genetic location of abs8752, between 8 and 10 o'clock, was similar to absA, at 10 o'clock, but antibiotic synthesis of abs8752 was similar to abs B's, at 5 o'clock. Abs8752 was complementated with multiple copies of the actII/ORF4 gene, an activator of synthesis of the antibiotic actinorhodin, and afsR, but not with absA. The xylE activity of reporter gene fusion to act structural gene cluster was not expressed in abs8752. This result suggests that abs8752 blocked transcriptional level like absA, absB. However xylE activity of reporter gene fusion to act structural gene was expressed in abs95. the result suggests that abs95 mutation is at the post transcriptional level.
