To develop yeast strains producing high concentration of ethanol from tapioca, screening and selecton from soil samples and mutation by gamma-irradiation were used. As the results, several yeast strains capable of producing higher concentration of ethanol than control yeast strain were obtained, where the control yeast strain is now being used in the production of ethanol in a Korean-ethanol manufactory. To develop yeast strains having cellulose-degradability, protoplast fusion and transformation techniques were used. Through the protoplast fusion between Saccharomyces diastaticus 1185 producing high concentration of ethanol and Brettanomyces curstersii H1-55 secreting β-glucosidase, I11 fusants having both of the parental chracteristics were obtained. Through transformation, p7 plasmid containing β-endoglucanase gene was introduced into Saccharomyces sp. K110, K114, K125, 1559 and their resulting transformants could secrete the β-endoglucanase into their culture media. Through the 'in vivo' mutation of K125 strain by using mutator E. coli, transformant Tm-4 showing higher β-glucanase activity and transformants Tm-6 and Tm-7 exhibiting lower 0 -glucanase activity were obtained.
