There has been few report about cellular response under 2 atmospheric pressure (ATA) pressure in spite of many researches have been done for cellular response from mechanical stimuli including ultra high pressure (over 100 ATA pressure) and low-level of high pressure (under 1.5 ATA pressure). The study for cellular response under 2 ATA could give new result in cellular response from mechanical stimuli. To study cellular response from 2 ATA pressure the custom- designed pressure cell incubator was made and the CO_(2) level and temperature of pressure cell incubator were well controlled for cell incubation. The morphology of Jurkat cells were compared by SEM between the cells incubated in normal incubator and pressure incubator. The surface of Jurkat cells incubated in pressure chamber showed irregular, suggesting 2 ATA pressure gave enough stimuli to change cell surface. We also compared the protein changes by 2 D electrophoresis between the cells incubated in normal incubator and pressure incubator. The protein sptos were scanned and analysed by Imagemaster and changes of proteins were confirmed by Maldi-Tof analysis. The total of 10 proteins were changed in Jurkat cells by 2 ATA pressure. Among them, Adenosine deaminase, Tropomyosin 4, Tubulin-specific chaperone A, Protein kinase C, Thioredoxin peroxidase 2, Rho GDP-sissociation inhibitor 2, Hydroxyacyl-coenzyme dehydrogenase, Esterase D, 26S proteasome decreased and only mitochondrial HSP(heat shock proein) increased in pressurized Jurkat cells comparing to Jurkat cells under normal 1 ATA pressure.
