Effects of citric acid on the extraction of polysaccharides from Sangwhang mushroom

    This study was performed to investigate the effect of citric and on the yields of extract and polysaccharides from Sangwhang mushroom, and the results are as follows. The extraction yield and the total sugar content increased with increase of extract temperature from 90℃ to 130℃. In distilled water extraction, the highest extraction yield was obtained at 120℃. In the case of extract by citric acid, the yield of extract increased with increase of the acid concentration. However, in the case of extract by hydrochloric acid solution, the yield of extract, total sugar and reducing sugar content were high, whereas the polysaccharides yield was very low. The optimum temperature and acid concentration for extraction were 120℃ and 5 mM citric acid, respectively. In the optimum condition, the extraction yield increased by 23.9% and the polysaccharides increased by 8.2% in comparison with extract by distilled water. The total sugar in extract increased by 8.3% and the polysaccharides increased by 17.9% in comparison with extract by distilled water. The polysaccharides obtained from distilled water extract and citric acid extract were comprised of galactose, glucose, mannose, and xylose. The main sugars were galactose and glucose and the composition of sugars in extract showed no big difference whether using distilled water or citricacid. The anti-complementary activity of extract increased with the citric acid concentration from 0.1mM to 5mM and the activity rather decreased by 10 mM citric acid and 10 mM and 100 mM hydrochloric acid. The anti-complementary activity of extract by 5 mM citric acid solution was 86.3% whereas the extract by distilled water was 54%.
