    Viroids are the smallest plant pathogens that cause serious damages to the crop plants including potato, tomato, hop, grapevine, and fruit trees such as apple, peach, pear, and plum. A rapid and sensitive method for detection of plant viroids using reverse transcription-polymerase chain reaction(RT-PCR) technique has been developed already. However, sometimes it is difficult to detect viroids from young plants, so it is necessary to develop the advanced diagnosis system for detection viroids. In this research, the low molecular weight RNAs containing viroid RNA molecular were extracted from young leaves, barks, and other tissues of apple trees which were infected by viroids, and were performed the experiment through RT-PCR and Nested-PCR assay. As shown by Nested-PCR, Apple Scar Skin Viroid-Korean strain(ASSVd-K) was detected more clearly than RT-PCR. The Nested-PCR assay for detection of viroids is a highly sensitive method as compared with RT-PCR assay to detect ASSVd-K from young trees during the early time of apple cultivation. It can be applicable to the prevention against spread of viroids.
