Selection and characterization of zebrafish nunborakr91 mutant displaying retinal degeneration

Large-scale genetic screens have been the most powerful approach for characterizing genes related to zygotic mutations affecting development. Through the mutagenesis, we have screened a nunbora^(kr91)(nun) mutant that displayed morphogenic defects in nervous system. In nun mutant, retina was degenerated with a loss of retinal ganglion commissure. To reveal the role of nun in retinal patterning, we analyzed the changes of gene expression for several markers related to the eye development. In 36 hpf nun embryos, expression of vax2 was reduced, while that of tbx5.1 was increased, suggesting that regulation of dorsal and ventral retina is differentially regulated by nun allelle. The establishment of neuronal cell types along the dorsoventral axis of the nervous system is mediated by Bmp, Fgfs and Hh signalings. Howere expression of these genes were not altered in nun mutant embryos at 24 hpf. To reveal the role of nun the brain patterning, I analyzed the expression for several markers related to the brain development. In 24hpf nun embryos, expression of emx1 markers telencephalon, and krox20 markers hindbrain, is indistinguishable, but expression of hlx1, zp50, lim3 and otx2 markers diencephalon and midbrain, is reduced. Taker together, these results suggest that allele nun plays an important role in dorsoventral patterning of retina and retinotectal patterning in the brain.
