Identification of Genotype of F9 and F10 Selections of Crosses for Introgression of Resistance to Phytophthora capsici into CMS-based Hybrid Breeding System with Respect to Fertility-restoring Gene : 역병저항성 웅성불임계 육성을 위한 F9와 F10세대의 회복계 유전자형 검정

선행연구에서 역병저항성 웅성불임계 육성을 위하여 유지계(Nrfrf, CMS-B)에 역병 저항성 재료인 AC2258(N(S)RfRf, CMS-C)과 SCM334(N(S)RfRf, CMS-C)를 교배하여 역병 저항성 검정은 F8세대까지, 임성검정은 F7세대까지 검정하였다. 본 실험은 F9∼F10세대의 역병저항성 검정과 F8∼F9세대의 세포질 웅성불임계의 임성을 검정하였다. 2003년도에는 CMS-A(Srfrf)와 F8세대를 교배하여 얻어진 F1 21개 조합을 이용하여 회복계 유전자형 검정을 하였다. 21개중 12개가 유지계(CMS-B)로 판명되었으며, 9개의 조합은 임성이 분리되는 현상을 보였다. F9세대는 역병 검정을 한 후에 역병저항성 웅성불임계 육성을 위하여 CMS-A에 여교배 하였다. 2004년도에는 F10세대의 역병 저항성 검정을 하였으며, 예상 되로 대부분의 계통에서 역병 저항성을 보였다. F9세대와 CMS-A를 여교배한 조합은 임성검정 뿐 아니라 역병 저항성 검정도 실시하였다. F9세대와 CMS-A를 여교배한 22개의 F1 조합에서는 임성이 확실히 구분되었으며, 유지계(CMS-B)가 10조합, 회복계(CMS-C)가 12조합으로 나타났다.
In the previous research, a maintainer (Nrfrf, CMS-B) was crossed with two sources of resistance to Phytoophthora capsci, AC2258 and SCM334, with restorer genotype (N(S)RfRf, CMS-C), and evaluation and selection for resistance was conducted thereafter to F8 generation and nuclear genotype was identified up to F7 selections. In this study, F9∼F10 generation was evaluated for resistance and the F8∼F9 selections were identified for their nuclear genotype interacting with the male sterile cytoplasm. In 2003, 21 F1 hybrids obtained by crossing CMS-A (Srfrf) with F8 selections were examined for male sterility for identification of the nuclear genotype of the pollen parents. Twelve of them revealed male sterility indicating that their polllen parents were maintainers (CMS-B) and nine were segregating in male fertility indicating that their pollent parents were heterozygous. The F9 lines were evaluated for resistance and selected individual plants were crossed with CMS-A lines again for identification of nuclear genotype or for introduction of Phytophthora resistance into CMS-A lines by backcross. In 2004, F10 selections were evaluated and selected for resistance to P. capsici. Almost all the F10 lines showed a high level of resistance to P. capsici as expected. The crosses between CMS-A and F9 selections, and the backcrosses of the male sterile F1's between CMS-A and F8 selections to their respective F9 selections with CMS-B genotype were examined for male sterility and were also tested for resistance to P. capsici. Male fertility was clearly distinguished. Of the 22 F9 selections crossed to CMS-A, 10 turned out to be maintainer (CMS-B) and 12 restorers (CMS-C). Thus, basic materials for breeding Phytophthora resistant hybrid cultivars are obtained.
