Selection of F_(2) genotype with lacking kunitz trypsin inhibitor and tagging of ti gene

Soybean Kunitz trypsin inhibitor is one of the well known antinutritional factor. Lipoxygenase has been implicated as a cause off-flavors in soybean oil products. This study was conducted to select F_(2)genotypes with lacking Kunitz trypsin inhibitor protein andlipoxygenase and to detect molecular marker linked to ti gene controlling Kunitztrypsin inhibitor absent using Near Isogenic Line. Total 90 F_(2) progenies were derived from the cross Jinpumkong2 and C242 to select F_(2) genotype with lacking Kunitztrypsin inhibitor and lipoxygenase. Also, total 56 F_(2) progenies were derived from the cross Gogteuk and C242 to select F_(2) genotype with lacking Kunitz trypsin inhibitor and black seed coat. Five seeds from each of the F_(2) plants in both populations were bulked and analysed to determine Kunitz trypsin inhibitor and lipoxygenase protein. The 21.5kDa band of Kunitz trypsin inhibitor protein was segregated in F population and 18 F_(2) genotypes with lacking Kunitz trypsin inhibitor protein were selected. Only one genotype with lacking Kunitz trypsin inhibitor and lipoxygenase was selected. Total 20 F_(2) lines were selected with black seed coat from the cross Gogteuk and C242. Four Flines with black seed coat and lacking Kunitz trypsin inhibitor were selected. Total 1,000 Operon primers were screened to identify molecular markers linked to ti gene. OPAX17 primer showed a polymorphic band between Clark(TiTi) and Clark-derived iVIL(TiTi). However, OPAX17 primer was not linked to ti gene in linkage analysis. Selected F_(2) genotypes in this study will be used to improve high quality soybean cultivars.
