    In the present study, the reverse transcription-polymerase chain reaction (RT-PCR) methods were determined to differentiate infections of transmissible gastroenteritis virus (TGEV), porcine epidemic diarrhea virus (PEDV) and rotavirus, which lead to much economic losses in the swine population in Korea. After the PCR conditions, such as primers and their annealing temperature, were confirmed, duplex or single PCR methods were established to diagnose and differentiate these virus infections. The RT-PCR methods were also applied to the field samples of piglets' intestines and feces to determine the efficacy. 1. For the differentiation of these viruses, proper and efficient PCR condition was determined as 30 cycles consisted of 1 minute each of denature at 94℃, annealing at 42℃ and polymerization at 72℃, with 1.0 mM MgCl_(2). 2. The duplex PCR was able to differentiate TGEV and PEDV by the detection of N gene, whereas it was unable to differentiate three viruses including rotavirus by triplex PCR, because of non-specific priming against rotavirus VP7 gene. 3. It was possible to undertake successful duplex PCR for the TGEV and PEDV, and single PCR for the rotavirus dually in a thermocycler with a primer-annealing temperature of 42℃. 4. By the established RT-PCR methods, it was possible to differentiate TGE, PED and rotavirus infection in piglets' intestines and fecal samples
