Detection of a novel single-stranded RNA mycovirus in pleurotus eryngiim

A novel mycovirus named Pleurotus eryngii Spherical Virus(PeSV) was isolated from cultivated Pleurotus eryngii (king oyster mushroom) with a severe epidermic after a purification procedure involving PEG-NaCl precipitation, differential centrifugation, and equilibrium centrifugation in CsCl(1, 7846g/㎤). PeSV was a 30∼32nm spherical virus, encapsidated a single-stranded RNA (ssRNA) of 7.8kb with a coat protein of approximately 22kDa. Nine monoclonal antibodies in mouse and polyclonal antibody in rabbit against the PeSV were produced. The monoclonal and polyclonal antibodies against the PeSV could not cross-reacted with other mushroom virus, OMSV, OMIV-Ⅰ, OMIV-Ⅱ, OMIV-Ⅲ and FvSV. The monoclonal and polyclonal antibodies developed in this study were successfully applied in Triple-antibody Sandwich ELISA(TAS-ELISA) for identification and detection of PeSV even though very small amount of virus harbored in fungal tissues. cDNA of PeSV genomic ssRNA was synthesized and partially sequenced. We used the partial nucleotide sequence as a specific primer to detect viral genome in P.eryngii by RT-PCR.
