Biosynthesis and biochemical characterization of a pyoverdine-like pigment in pseudomonas fluorescens BM07

P. fluorescens BM07 cells grown on 70 mM succinic acid secreted a yellow-green pigment to culture medium. The pigment was purified using XAD-4 chromatography followed by CM Sephadex chromatography. The isolated pigment was characterized by amino acid analysis, UV/Vis spectroscopy, MALDI-TOF, Q-TOF and NMR spectroscopy. It was temporally identified as pyoverdine. The stoicheiometry of the Fe(Ⅲ)-pigment complex was 1:1. The spectral characteristics of Fe(Ⅲ)-chelated pyoverdine depended on pH. The pigment was considered to belong to an atypical pyoverdine that has two β-OH-Asp and chromophore as iron-binding ligands. The pigment can chelate Fe(Ⅱ) as well as Fe(Ⅲ). The amino acid composition of this siderophore was Serine, Glycine, Alanine and Lysine (1:2:2:3). Its molecular weight was about 1300. It was found that while pyoverdine was secreted to culture medium, little PHA was accumulated in the cell. When P. fluorescens was grown on fructose plus succinic acid, pyoverdine production was more favored than cell growth depending on succinic acid concentration. It is considered that there must be some genes linked between pyoverdine and PHA production pathway.
