Backgrounds : Vitiligo is caused by the destruction of melanocytes. While the precise cause of melanocyte destruction is unknown, potential pathologic mechanisms include neural, self-destruction, autoimmune hypothesis. Recently, abnormalities of both humoral and cell-mediate immunity have been well documented in patient with this disorder, which provided theorical base for introducing tacrolimus ointment, working as an immunomodulator, in the treatment of vitiligo. Objectives : This study was performed to examine the effects of tacrolimus on the survival rates of keratinocytes and melanocytes of vitiligo and on the melanization of melanocytes. Methods : Keratinocytes and melanocytes were prepared and cultured from normal skin of 4 vitiligo patients. They were treated with tacrolimus of various concentrations (0.01, 0.1, 1, 10, 100 μM) for 5days. Thereafter, MTT(3-(4,5-cimethylthiazol-2-yl)-2,5-di-phenyl tetrazolium bromide) assay and melanin content analysis were done. Results : Tacrolimus showed no significant effects on the survival rates of keratinocytes. However mild proliferative effects of melanocytes were shown in the concentration of 10 μM, on the other hand, marked cytotoxicity was observed over the concentration of 100 μM, and no effect under the concentration of 1 μM. The melanin contents of individual melanocytes were decreased according to the concentration from control to 10μM. Conclusion : These findings suggested that tacrolimus did not have proliferative effects on keratinocytes and melanocytes of vitiligo patients, and had tendency to decrease the melanin contents in each melanocyte.
