Visceral pain is a tremendous clinical problem but knowledge of specific mechanisms underlying visceral pain has lagged due to confusion about adequate stimuli and the lack of useful animal model. Recently a rat model of cyclophosphamide (CP) cystitis was developed that requires only intraperitoneal injection and features inflammation confined to the bladder. However, there is a possibility that CP may influence the uterine tissue, considering a case report that CP-treated woman developed uterine inflammation. So, this study examined the extend of inflammatory response, the expression of substance P following intraperitoneal injection of CP. As results, there were no changes in plasma protein extravasation and inflammatory response of uterine tissue, though there were increased responses of plasma protein extravasation and inflammatory response in urinary bladder tissue. In the uterine tissue, however, SP immunoreactivity increased 8, 12h after CP injection. These results suggest that CP-induced cystitis rat model is appropriate and the increased response of hypogastric or pelvic nerve resulting from cystitis by CP injection may influence SP expression of uterine tissue.
