    Background. Lung cancer is a leading cause of cancer death in Korea and rapidly increased it's prevalence. Single nucleotide polymorphism (SNPs) represent an important class of genetic variation and susceptibility of individual. Recently, many studies have been done regarding SNPs of indicator gene for lung cancer. But no study was done for Korean specific SNP of lung cancer. To search Korean SNP for lung cancer, we analyzes the frequency of SNPs related lung cancer in Korean Lung cancer patients and normal population. Methods. In first study included 50 Korean lung cancer patients and 43 normal population. We analyzed frequency of known SNPs related to lung cancer by SNaPshot assay in 99 site of 30 gene. We identified Korean lung cancer related SNP site at 10 points from 99 points, and the related site was followed; RAGE gene (-6277 T/A), MMP1 gene (I53V), MTHFR gene (A222V), TSSC5 gene (A1080G), DLK1 gene (V104M0, FHIT gene (H98H), FUS2 gene (A875T), LOH11CR2A gene (H386H), MMP2 gene (-1306C/T) and USP25 gene (A601A). In second comparative study with 182 lung cancer patients and 192 normal population could determine precise lung cancer risk in 10 SNP point. Results The frequency of MTHFR A222V C/T polymorphism was significantly different between lung cancer and control group. The odds ratio of risk of the lung cancer with the MTHFR (A222V) C/T polymorphism was 0.555 (p=0.031) and LOH11CR2A T/C polymorphism was 1.740 (p=0.0175). In MDR analysis it does not show a significant gene-gene interaction. However, adding age, smoking, and gender to the MDR analysis reveals a highly significant interaction between age and smoking (prediction error = 19.73%, P
