    Bioassay has been tried to determine dioxins in food. The recombinant cell line from human hepatoma HepG2 cells is modified to include a luciferase reporter plasmid responsive to the liganded AhR­Arnt complex. When dioxins are present in the cells, they produce luciferase, which oxidize the added substrate luciferin and the emitted luminescence is detected by a luminometer. DRE­Luc cells were maintained in DMEM with 10% fetal calf serum at 37℃ in a humidified CO_(2) controlled incubator. The food samples were extracted in dichloromethane and the extract was cleaned­up using by sulfuric acid impregnated silica gel, re­dissolved in DMSO and applied to bioassay. In addition, reconsider analytical effect generated by bioassay have been compared to the assay results of HRGC/HRMS and bioassay. As results, to measurement luciferase activity was required 24 hours exposure time and able to analysis in 3∼1600 pg/g quantitative range. The average recoveries were 108% and the levels of dioxins in meat samples were below 30 pg/g.
