Background : Receptor tyrosine kinases(RTKs) play critical role in normal development, growth and tumorigenesis in human tissue. The purpose of this study is to evaluate the effect of RTK in progression of breast cancer. Methods : The expression of EphA2, c-met, and c-erbB-2 was examined in breast tissue including 111 normal epithelium, 34 ductal carcinoma in situ(DCIS) and 109 invasive ductal carcinomas(IDC) using immunohistochemical method and RT-PCR. The results were compared with the prognostic parameters of breast cancer including tumor grade, growth pattern, tumor size, lymph node metastasis, and the expression of ER, PR, p53 and ki-67. Results : The protein expression of three RTKs was higher in DCIS and IDC than in normal epithelium. The protein expression of EphA2 was correlated with high grade tumor and high labelling index of ki-67. Also it was correlated with the protein expression of c-met in IDC. The overexpression of c-erbB-2 was correlated with lymph node metastasis. The mRNA expression of three RTKs was higher in low grade tumor and was closely correlated with each other in normal tissue and IDC, respectively. Conclusion : Three RTKs have critical role in tumorigenesis in human breast cancer. The overexpression of EphA2 may be a poor prognostic parameter of breast cancer.
