Fruits of Opuntia ficus-indica is known to have beneficial effects such as antiulcerogenic and antioxidant activities. In this study, Fruits of Opuntia ficus-indica were hydrolyzed by enzyme, then, fermented with two species of yeast. And we identified the new distinguished markers between EO (enzymatic hydrolyzed Opuntia ficus-indica) and FO (fermented Opuntia ficus-indica) samples using UPLC-QTOF-MS/MS and UHPLC-LTQ-Orbitrap and evaluated protective effect of EO and FO samples on photoaging in the skin cell culture and hairless mice with UV irradiation. It was found that major fermented metabolites in FO sample were caffeic acid, p-coumaric acid, ferulic acid, isorhamnetin 3-O-rutinoside and isorhamnetin 3-O-glucoside. And, treatment of FO sample in skin fibroblast with UVB irradiation increased the TGF-β1 production. In the HaCaT keratinocyte and NHDF fibroblasts coculture, treatment of FO sample significantly decreased in IL-1β, MMP-1 and TNF-α production. An in vivo study using hairless mice showed that topical application of FO sample protected mouse skin from photoaging. Thus, the present study demonstrated that fermented Opuntia ficus-indica (FO) functions as an anti-skin aging against UVB irradiation-induced photo damage to skin.
