    This study was conducted to determine the germinability according to the temperatures, to establish the methods of seed pre-treatment for improvement of seed germination, and to understand the proteomic changes during the germination in spinach cultivars with different seed morphological character, such as round and prickly type. Results are summarized as follows. 1. Effect of temperature on germinability of spinach seeds Seed germinability such as germination rate, T50 and MGT in spinach seeds were markedly influenced by the temperatures. Round seeds of spinach germinated much fast than prickly seeds at 5, 10, 15 and 20oC. When germinated at 25oC, prickly seeds failed to germinated as 11%, whereas round seeds showed about 51% germination rate. Among the tested temperature regimes, best seed germinability of round and prickly seeds was obtained at 15 and 10oC, respectively. 2. Effect of seed pre-treatment on germinability of spinach seeds Priming treatment increased germination rate and reduced T50 (days for 50% of the final germination) and MGT (mean germination time) in round and prickly seeds. All priming treatments with PEG, Ca(NO3)2, K3PO4 or KNO3 led to a significantly more rapid germination compared to that of nonprimed round seeds when germinated at 5 and 15oC. At 25oC, such an effect of seed priming was decreased. When germinated at 5 and 15oC, germination rate of prickly seeds primed with Ca(NO3)2 and K3PO4 was increased and reduced T50 and MGT values compared to that of nonprimed seeds. However, all priming treatments of prickly seeds failed to germinated up to 50% at 25oC. Seed pre-treatment with plant growth activators such as ALA, GB and JA increased germination rate and reduced T50 and MGT in round and prickly seeds when germinated at 5 and 15oC. At 25oC, all treated prickly seeds failed to germinate up to 50% whereas round seeds showed about 68~95% germination rate according to plant growth activators. 3. Proteomic changes during germination of spinach seeds The proteome profiles of round and prickly seeds were compared on 2-dimensional gels. More than 300 protein spots in fresh seeds were reproducibly detected by two-dimensional gel electrophoresis. when germinated at 5 and 15oC, reproducible protein spots were increased compared to that of fresh seeds in round and prickly seeds. At 25oC, reproducible protein spots decreased compared to that of fresh seeds in both cultivar. The differentially expressed proteins spots between round and prickly seeds were selected from the total protein spots. The 24 spots were excised from the gel and subjected to further analysis. Twenty two spots out of 24 spots were identified from spinach and Hibiscus tiliaceus, whereas 2 spots were unidentified.
