프루텔고치벌(Cotesia plutellae) 독샘 전사체 분석과 면역관련 Calreticulin 유전자의 특성 구명

As endoparasitoid wasp, Cotesia plutellae is natural enemy against the diamondback moth., Plutella xylostella via laying egg into larvae. For successful parasitism of the offsprings, the female wasp injects maternal factor such as polydnavirus, venom and ovary proteins. These maternal factors protect their eggs from host immune system as well as modulate their host physiologically. In order to analyze the function of immune suppression of venom, transcriptome analysis of the venom gland was performed. As a result of analysis of transcriptome, 57 genes were related to the immune responses such as calreticulin, arrestin, integrin, defensin etc. In order to identify of immunosuppressive activity of calreticulin preferentially, we obtained calreticulin via performing molecular cloning. Calreticulin was functionally expressed in Escherichia coli via SDS-PAGE and Western blot. Expressed calreticulin was purified using his-taq and injected into the host to examine if it is involved in host immune suppression. When injected into the host, suppression of nodule purified calreticulin exhibited suppression of nodule formation. These results suggest that venom gland inhibits immune response of host, and calreticulin is related with suppression of cellular immune response in the host.
