A Study on the Effect of Porous Nerve Guide Conduit with Growth Factor Gradient on Peripheral Nerve Regeneration

we prepared an asymmetrically porous polycaprolactone (PCL)/Pluronic F127 membrane with NGF concentration gradient. The long gap (2cm) nerve guide conduit (NGC) was fabricated by rolling the membrane. It was observed that the PCL/F127 asymmetrically porous membrane has the gradually increasing concentration of NGF. The NGF immobilized on the asymmetrically porous membrane was released in a sustained manner for up to 28 days. The proliferation /migration of nerve cells (via PC 12 cell culture) and nerve regeneration/functional recovery (via animal study) were more effective for the NGC immobilized with NGF concentration gradient than the NGC with uniform NGC concentration. From the results, we suggest that the NGC with asymmetrically porous structure and concentration gradient of NGF can be a simple and effective therapeutic technique to accelerate the reinnervation rate and provide sufficient functional recovery of peripheral nerves.
