In this study, a novel CDs production process was designed and optimized using highly soluble sucrose as an initial raw material. Cyclodextrin glucanotransferase (CGTase) was used to produce CDs from the linear α-(1,4)-glucan that was supplied by amylosucrase (NpAS) treatment on sucrose. The maximum CDs yield (21.1%, w/w) was achieved from simultaneous dual enzyme reaction mode which was 8.4 U/mL NpAS and 0.75 U/mL CGTase treatment at 40°C for 24 h. . The maximum production of CDs (15.1 mg/mL) with 0.5 M sucrose was achieved with 8.4 U/mL NpAS and 3.75 U/mL CGTase at 40°C for 24 h in a simultaneous dual enzyme reaction mode. From the CDs production by the sequential enzyme treatment, the yield of α-(1, 4)-glucans ranged from 19.6 to 29.8% at 0.1 to 1.0 M sucrose. Also the yield of CDs was 29.0 to 85.1% depending on the size of α-(1, 4)-glucans. Consequently, simultaneous synthesis of CDs was successfully carried out by this dual enzyme system without starch pre-treatment steps.
