Antioxidant and Anti-diabetic Activities of Various Solvent Extracts from Boletopsis leucomelas FAYOD

    The aim of this study is to determine the antioxidant activities and in vivo activity on STZ-induced animal model by extracts of Boletopsis leucomelas FAYOD. B. leucomelas, an edible mushroom, has a bitter taste and burnt odor with a black fruiting body. The dried B. leucomelas was extracted with distilled water, 100% ethanol and 100% methanol. The extraction yields (%) were 35.53, 10.16, 25.96 by D.W., 100% EtOH, 100% MeOH, respectively. Total phenol and flavonoid contents of 100% EtOH extracts were higher compared to other extracts at 965.14±31.53 mg gallic acid equivalents (GAE)/g, 815.01±35.21 mg catechin equivalents (CE)/g, respectively. The highest antioxidant activity was significantly found on 100% MeOH extracts for DPPH radical scavenging activity, ferric reducing antioxidant power (FRAP), cupric ion reducing antioxidant capacity (CUPRAC), while, the oxygen radical absorbance capacity (ORAC) was significantly high in the 100% EtOH extracts. The α-glucosidase inhibitory activity of 100% EtOH and 100% MeOH extracts were significantly higher than D.W. extracts. In oral glucose tolerance test (OGTT), twenty BALB/c mice were divided into four groups which was no treatment group, STZ-induced diabetic mice, fed with 50 mg/kg of acarbose in STZ-induced diabetic mice, fed with 200 mg/kg of 100% EtOH extracts powder from B. leucomelas dissolved in D.W. in STZ-induced diabetic mice. Consequently, the glucose level of B. leucomelas extracts treated group was significantly decreased compared to that of the acarbose treated group. In conclusion, 100% EtOH and 100% MeOH extracts of B. leucomelas will be useful as natural antioxidant and potential materials for anti-diabetes therapeutic or preventive therapy.
