    In vitro plantlets of eight potato varieties 'Superior', 'Gogu Valley', 'Juice Valley', 'Early Valley', 'Winter Valley', 'Taedong Valley', 'Purple Valley', and 'Golden Valley' were used to investigate the effect of an additional Murashige and Skoog (MS) liquid medium supplemented with sucrose and plant growth substances (PGSs) on the production of large size microtubers. Plantlets were grown in MS solid medium containing 6% sucrose and incubated for the first 60 days under long day condition (16 h light/day), and then added with MS liquid medium with 8% sucrose. These plantlets produced the highest numbers (1.7 microtubers/plantlet) and yield (635 ㎎/plantlet) of microtuber. The lowest number and yield were obtained from the in vitro plantlets grown in the solid medium without adding MS liquid medium. As for the utilization of PGSs, it was shown that the higher the PGSs levels, earlier the tuberizations with higher yield and lower number of microtubers. The highest yield (591 ㎎/plantlet) was obtained from 200 ㎎ㆍL⁻¹ of succinic acid 2,2-dimethylhydrazide (B-9) and numbers (1.6 microtubers/plantlet) from 10 ㎎ㆍL⁻¹ of 6-benzylaminopurine (BAP) treatment. In the case of combinations of PGSs, treatments of 10 ㎎ㆍL⁻¹ BAP and 200 ㎎ㆍL⁻¹ B-9 produced significantly the highest numbers (1.5/plantlet) and the highest yield (766 ㎎/plantlet) of microtuber, but lowest from the control. Thus, an extra addition of MS liquid medium with sucrose and PGSs might have played a major role for the further induction and development of large size of microtubers on diverse genetic background of potatoes under in vitro conditions.

    The anthers of tobacco (Nicotiana tabacum cv. Bright Yellow-4) were irradiated with ¹⁴N-heavy ion beam at 5 Gy, and then haploids were selected in M₁ generation (pale-pink flower color). The haploids of M₁ generation were self-pollinated and flower color mutants were selected from M₂ generation. The phenotypic and molecular differences were observed between the mutants and the non-mutants. The petals of the mutants were rounder in shape and had much lighter color compared to those of non-mutated ones. The dot blotting and Northern hybridization analysis revealed that the chalcone synthase (CHS) gene in flower tissues was less expressed in the mutants than in the non-mutants. This implies that the expression of CHS gene in the anthocyanin biosynthesis pathway could be suppressed in mutant plants.

    This study was conducted to compare the growth and fruit characteristics of diploidy (2N = 22), triploidy (3N = 33), and tetraploidy (4N = 44) 'Mudeungsan' watermelon (Citrullus vulgaris). Seedlings and rooted cuttings of diploid grew earlier than those of triploid and tetraploid in the early growth stage. The higher the ploidy level, the larger the plant organs in general. The conspicuous differences among diploid, triploid, and tetraploid were on leaf length, width, area, and chlorophyll content, and tendril size. The DNA content per cell measured by flowcytometry was 1.5- and 2-times higher in triploid and in tetraploid, respectively, than that in diploid. Diploid bore the first male flower at lower node than triploid and tetraploid. There was only a little difference in fruit set between diploid selfings and triploid pollinated with diploid. Fruit set of tetraploid was very low at 12.5% when selfed, while when pollinated with diploid it was comparatively high at 60%. The width/length ratio of fruits from diploid selfings, from triploid pollinated with diploid, and from tetraploid pollinated with diploid was 0.84, 0.92, and 1.02, respectively. The self-pollinated tetraploid fruits were mostly malformed and relatively small. The fruits from diploid selfings contained about 380 seeds per fruit, while those from triploid pollinated with diploid contained about 162 embryoless, but tough seed coats.

    Pepper anthracnose caused by Colletotrichum gloeosporioides and C. acutatum is the most destructive disease affecting the Capsicum genus, causing serious yield loss and quality deterioration in many Asian countries including Korea. C. annuum is the major cultivated species worldwide, but many attempts to find resistant resources to the disease have been failed during the last two decades. A total of 326 pepper accessions within five domesticated Capsicum species were screened using a microinjection method to identify genetic resources for anthracnose resistance. Only 12 accessions belonging to C. baccatum displayed low disease incidence. Among them, PBC80 and PBC81 were selected to be the highest resistant lines and used to more-thoroughly examine their resistant properties for resistance breeding. Both lines were comparatively resistant to Colletotrichum pathogens isolated from Taiwan and Indonesia, as well as from Korea. In addition, their strong resistance was well maintained even through natural field infections in Korea, Indonesia, and India. Both accessions seemed to be reliable and usable genetic resources for anthracnose resistance and could be used to develop resistant varieties of chili pepper if we can use interspecific hybridization.
