  1. [국내논문]   Genetic Divergence and Speciation of Eurasian and American Yellow Perch Based on the Nucleotide Sequence of Cytochrome b Gene

    Eurasian yellow perch (Perca fluviatilis) and American yellow perch (Perca flavescens) are known to be endemic species in Eurasia and North America, respectively. The presence of endemic species on each continent suggests their independent evolutionary history. However, because of the morphological similarity, distribution pattern, and only recent fossil record, their divergence time and speciation of the two Perca species has long been controversial. Here, from the comparison of the entire nucleotide sequences of cytochrome b gene, large genetic divergence between the two Perca species is observed although they are morphologically similar each other. Among 1,140 base pairs, interspecific nucleotide differences are found at 130 sites $(11.4\%)$ . The differences varies with codon position, showing 22 sites in the first, 5 sites in the second, and 103 sites in the third codon position. Considering the types of nucleotide changes, transitional differences are much more than transversional differences and its ratio turned out to be 5.19. The estimated divergence time of the two Perca species indicates that they were separated each other approximately in the late Miocene period, which implies the long history of speciation. With comparison of the inferred amino acid sequences, strong structural and functional constraints which seem to be maintained by the highly conservative amino acid residues or protein regions, as found in other taxonomic groups of organisms, are also recognized in the cytochrome b of the fishes examined.

    This study is to estimate abundance of the Chesapeake Bay blue crab stock. Using 823 dredge tows which were conducted during the 1991 winter survey, blue crab abundance was estimated on the basis of newly developed methods which account for unequal dredge tow distances, size- and sex-specific heterogeneous overwintering spatial distributions, wintertime exploitation, the time period of quasi-hibernation, and dredge capture efficiency. The estimate of total abundance before correction by gear efficiency was 131.8 million crabs $(95\%\;C.I.\;=\;118.2\;million\;crabs\;to\;145.4\;million\;crabs),$ Dredge capture efficiency was estimated to be 0.474. Thus, the estimate of total abundance was calculated as 278.1 million crabs after correction by the efficiency factor.

    초록

    A Monte Carlo computer simulation study was conducted to determine the most efficient sampling design for the blue crab dredge capture efficiency experiment performed in Chesapeake Bay, Maryland, U. S. A. The input values were the number of dredge tracks in each experimental area, the number of tows per experiment, the number of experiments, the mean density of crabs per unit area, the negative binomial coefficient, the gear capture efficiency, and the tow error. As a result of the study, a four-track experiment with twenty to twenty-eight tows was estimated to be the best in terms of precision and accuracy of the gear capture efficiency.

    The peptidyl prolyl cis-trans isomerase(PPlase, EC 5.2.2.8) from Bacillus stearothermophilus SIC1 was extracted from the cells treated with by lysozyme. PPlase was purified from the cell extracts by heat treatment, ammonium sulfate precipitation, ion exchange chromatography and finally gel filtration (FPLC). The purity of purified the enzyme after Superose 12 column chromatography was examined by sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE). The molecular weight of the purified PPlase was estimated as 18,000 by SDS-PAGE. The 39 amino acid residues from the N-terminus were determined by the protein sequencer. The enzyme showed the optimum pH at 8.0 and was stable at the range of pH 7.0 to 8.0. The enzyme was considerably stable after heat treatment at $60^{\circ}C$ for 30 minutes, and the enzyme was quite stable up to $65^{\circ}C$ . The presence of the PPlase in the refolding solution accelerated the isomerization rate of the assay peptide.

    Fine structural changes of germ cell during the gametogenesis of Korean rockfish, Sebastes schlegeli sampled in west coast of Korea were investigated from September 1993 to August 1994. In a layer of microvilli of oocyte with active yolk duplication, many pinocytotic vesicles containing protein granules regarded as yolk precursors were observed. The multivesicular bodies were formed by gathered mitochondria. They are participated in formation of the primary yolk globules homogeneously filled with high dense particles and enclosed within a limiting membrane. The precursors of yolk globule appeared to be formed by modification of mitochondria and they developed into the primary yolk globules with participation of large and dense pinocytotic vesicles. Yolk globules in mature oocyte were consisted of three components: the crystalline type main body, the superficial layer with dense and fine granules, and the limiting membrane. Steroid hormone secreting cells were recognized in the interstitial cells of growing testis. Numerous endoplasmic reticula and large mitochondria with well developed tubular cristae appeared in their cytoplasms. The axoneme in the tail flagellum of spermatozoon consisted of nine pairs of microtubules at the periphery and one pair at the center, and they were covered with doublet microtubules.

    This study was conducted to determine the serum vitellogenin (VTG) concentration changes during the ovulation period in elkhorn sculpin, Alcichthys alcicornis. The results of sepacryl S-300 showed that the molecular weight of VTG could be 380,000. SDS-polyacrylamide gel electrophoresis (SDS-PAGE) analysis may indicate that the purified VTG consists of three subunits with molecular weights of 180,000, 118,000 and 85,000, respectively. Yolk protein purified from the egg extracts was eluted on an equilibrated sephacryl S-300 column, and its molecular weight was estimated 250,000. The precipitation lines of the female serum against the antiserum of the egg extracts were fused completely by immunoelectrophoresis and immunodiffusion analysis. VTG was detected in the serum, and hepatocytes from males injected with $17\beta-estradiol\;(E_2).$ Furthermore, VTG was immunochemically similar to yolk proteins. The concentration of VTG was high before ovulation $(9.80\pm0.81-11.02\pm0.09 mg/ml),$ and then decreased rapidly after ovulation $(less\;than\;6.19\pm0.59 mg/ml).$ This study suggested that VTG was synthesized in the liver by the action of $E_2$ and released to blood, and then incorporated into oocytes.

    About 2.1g of pale yellow flavor concentrate was obtained from 10kg of chopped fresh ascidians through a Likens-Nickerson steam distilllation/solvent extraction. These concentrates could be fractionated to neutral $(91.5\%),\;basic\;(1.0\%),\;phenolic\;(3.2\%),\;and\;acidic\;(4.3\%)$ fractions. Total 65 volatile compounds were identified from those concentrates. The neutral fraction was representative flavor fraction which showed a similar flavor of total steam distillates of ascidian. The major compounds $(38.2\%\;of\;neutral\;fraction)$ were identified as carbon atoms 8 to 10 of alcohols. Among these volatile alcohols, 1-octanol, 2,7-decadien-1-o1, 3-octen-l-01, 7-decen-l-ol, and l-decanol were the dominent compounds found in neutral fraction. But the basic, phenolic, and acidic fractions differs from ascidian steam distillates flavor.

    Anchovy Engraulis japonica boiled and dried was stored at $20^{\circ}C$ for 5 months after that treatment of sodium-erythorbate (Na-ery) or deoxygenizer (Deoxy). During storage, peroxide value (POV), thiobarbituric acid (TBA) value, lipid content, and lipid class compositions were determined to evaluate the quality of the samples. pay was decreased rapidly for the first 3 months storage and its decrease was Deoxy group>Control group>Na-ery group in that order. TBA values increased for the first 4 months and then decreased rapidly, and it's increase was the highest in Control group, followed by Na-ery and Deoxy group. Total lipid contents in all samples declined during storage. Especially, phospholipid decreased mainly in Na- ery and Deoxy group, while neutral lipid mainly in Control group. Triglyceride (TG), phosphatidylethanolamine(PE), and phosphatidylcholine(PC) decreased, while free fatty acid (FFA) and lyso-PC (LPC) increased during storage. The decrease of TG was the highest in Control group and that of PE and PC was higher in Na-ery group than in other sample. The decrease of PE in all samples (except Deoxy group) was higher than that of pc. The increase of FFA and LPC were higher in Control and Na-ery group than in Deoxy group. These results indicated that the lipid deterioration of the boiled and dried-anchovy was effectively suppressed by the enclosed deoxygenizer during storage at $20^{\circ}C$ .

    The effects of deoxygenizer (Deoxy) and sodium-erythorbate (Na-ery) on the changes in fatty acid compositions were investigated to prevent the loss of n-3 polyunsaturated fatty acids in lipid of bolied and dried-anchovy during storage. After storage for 5 months, docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) of total lipid (TL) in Deoxy group decreased only $3.0\%\;and\;0.5\%$ , respectively, compared to those of before storage. However, those in Control group decreased 9.5\%\;and\;2.3\%,$ respectively. In the case of Na­ery group, the percentages of DHA and EPA decreased were lower than those of Control. Most of DHA and EPA in n was lost in the early stage of storage. Total DHA remained in phospholipid (PL), triglyceride (TG), and free fatty acid (FFA) fractions after storage for 5 months were $98\%,\;66\%\;and\;62\%$ in Deoxy, Na-ery and Control group, respectively, and total EPA remained was slightly high level compared with those of DHA. The loss of DHA was more in PL than in TG. Consequently, deoxygenizer was recognized as a good material to prevent the loss of DHA and EPA of the boiled and dried-anchovy during storage.

    Polychaetous community survey in Tomioka Bay was carried out 5 times seasonally from May 1991 to March 1992 by quantitative grab sampling (0.05m2) at 11 stations. Based on the granulometric composition and environmental factors, a homogeneous soft bottom was found in St.5-10. The species of the polychaete were classified into three feeding groups using the Fauchald and Jumars' feeding guild system. According to polychaetous community composition data, deposit feeders predominate in sandy silt area where the silt-clay content is $60-69.3\%.$ These deposit feeders were subdivided into surface deposit feeders and subsurface deposit feeders by their living position and mode. Also, suspension feeding group comes as the third dominant group. Seasonal changes of each feeding group were described in terms of numerical density and biomass. Feeding layer and types of dominant species (Lumbrineris longifolia: surface deposit feeder; Praxillella pacifica: subsurface deposit feeder; Chone duneri; suspension filter feeder, etc.) were examined in the intact sediment core samples. Also, longterm density change among the three dominant species during 10 years was disussed.
