    The effects of pH, temperature and various inhibitors on the proteolytic activity of the cell lysate of Uronema marium were investigated using colorimetric and substrate gel electro­phoretic methods. The cell lysate of U. marinum showed proteolytic activity over a wide range of pH, and pH optima ranged from pH 5 to 7. The proteolytic activity was increased according to a rise of temperature but decreased at $40^{\circ}$ . The proteolytic activity of the parasite lysate was significantly inhibited by protease inhibitors including trans-epoxysuccinyl -L-leucylamido-(4-guanidino) butane (E-64), pepstatin A, phenyl-methanesulfonyl fluoride(PMSF), and ethylenediamine-tetraacetic acid (EDTA). Preincubation of the lysate with E-64 showed the maximum inhibition of the caseionolytic activity. Four protease bands (152, 97, 67 and 40 kDa) were detected by gelatin SDS-PAGE. Significant inhibition of caseinolytic activity and complete abolition of a 152 kDa band in gelatin SDS-PAGE by EDTA indicated that the cell lysate of U. marinum had a metalloprotease Another three proteolytic bands were inhibited by E64, a cysteine protease inhibitor. Preincubation of the cell lysate with pepstatin or PMSF had no effects on the protease bands.

    Marine bacteria producing algicidal substance against Cochlodinium polykrikoides was screened and isolated from seawater. Metabolite of marine bacteria Micrococcus sp. LG-5 showed strong algicidal activity against C. polykrikoides. C. polykrikoides was inhibited above $90\%$ at $5\%$ solution of the metabolite within 24 hrs. Algicidal substance from the metabolite was extracted with ethyl acetate, and then purified by PTLC and reversed-phase HPLC. Algicidal activity of purified compound against C. polykrikoides was above $90\%$ at 3.7, 11.0 and 33.0 ${\mu}g/mL$ concentration after 12, 9 and 3 hrs, respectively. Ninety percent inhibition of other red tides, Gymnodinium sanguineum and Gyrodinium impudicum was observed when treated with 3.7 ${\mu}g/mL$ of purified compound within a period of 12 hrs. The microscopic view of red tides treated with purified compound showed the deformations such as cell node cuts and swelling of cells.

    Enteromorpha prolifera of the isomorphic diploid sporophyte and the haploid gametophyte generations inhabit rocks, tidal flats and tidal pools in the middle parts of intertidal zones. In this experiment, their thalli were observed by bare eyes from October and experienced $74\pm16.5cm$ maximum growth the following March and April. The rate of occurrence of the thalli per month was highest in March, while their biomass peaked at $1,464\pm41.5 g/m^2$ in Jangheung in April. Genetic similarity was investigated samples of E. prolifera collected from Muan, Wando, Jangheung, Yosu and Jinhae, at the south coast of Korea. Random amplified polymorphic DNA (RAPD) markers were used. For the RAPD analysis, 3 ng of the DNA extracted from the thalli using he phenol/chloroform method was amplified by PCR with a 25 {\mu}L$ reaction solution, arbitrary primers and 36 cycles. Among the 60 primers used, 31 yielded products, most of which showed diverse electrophoresis patterns. Similarities among the groups compared ranged from 0.37 to 0.58. We conclude that the use of RAPD analysis is appropriate to characterize the genetic variability of this commercial species along its geographical distribution.

    This study was conducted to investigate the effects of dietary lipid sources on survival, growth anc body composition of snail (Semisu1cospira gottschei). Three replicate groups of snail (average weighing 152mg) were fed the diets containing different lipid sources such as lauric acid (LA), squid liver oil (SO), linseed oil (LO), corn oil (CO), SO+ LO, SO+CO, LO+CO and SO+LO+CO for 8 weeks. Survival was not affected by dietary lipid sources (p>0.05). Weight gain of snail fed the SO, SO+LO and SO+LO+CO diets was significantly higher than that of snail fed the LA and LO diets (P 0.005). Contents of 12:0, 18:2n­6, 18:3n-3 and 20:5n-3 from snail fed the LA, CO, LO and SO diets were higher than those from snail fed the other diets, respectively (p 0.05). The n-6 highly unsaturated fatty acids such as 20:4n-6 and 22:4n-6 contents of snail were not affected by dietary lipid sources (p>0.05). These results suggested that squid liver oil and mixture of squid liver oil and linseed and/or corn oil are good dietary lipid sources for the normal growth of snail. However lauric acid may not be a good lipid source for snail diet.

    This study was conducted to investigate the utilization of Sargassum meal in the diet on juvenile ayu (Plecoglossus altivelis) reared in seawater. White fish meal and wheat flour were used as the dietary protein and carbohydrate sources in the control diet. Wheat flour in the control diet was replaced with $5\%$ and $10\%$ Sargassum meal. Three replicate groups of fish average weighing 4.0 g were fed one of three isonitrogenous $(45\%)$ and isocaloric (14.5 MJ /kg diet) diets for 7 weeks. Survival of all groups were above $80\%$ . Weight gain, feed efficiency and protein efficiency ratio were not significantly affected by dietary Sargassum meal levels (P>0.05). There were no significant differences (P>0.05) in moisture, crude protein, crude lipid, crude ash and fatty acid compositions of whole-body fish among groups. It is concluded that Sargassum meal could be used as a dietary additive or alternative low-cost dietary ingredient up to $10\%$ for juvenile ayu reared in seawater.

    The authors identified 68 Vibrio strains from Gwangan beach seawater from June to October in 2001. We identified them as 19 strains of Vibrio alginolyticus, 15 strains of V. vulnificus, 15 strains of V. parahaemolyticus, 11 strains of V. cholerae non O1, 7 strains of V. fluvialis and just one strain of V. hollisae. They showed their typical biochemical characteristics by API 20E kit (bioMerieux), respectively. It was examined whether their cultural supernatants had enzymatic activities such as hemolysin, protease or urease. The 46 strains showed hemolytic activities and/or protease activities. But we could not find any strain which had urease activity. All isolates of V. cholerae non O1 showed $\beta$ hemolysis. The others showed $\alpha$ hemolysis or did not show clear zones on sheep blood agar plates. These results of Kanagawa phenomenon were not always correspondant with hemolytic activities of cultural supernatants at late log phase. Some strains had higher hemolytic activities despite of showing protease activities on skim milk agar plates and in litmus milk media. On the other hand, some strains showed protease activities but did not show hemolytic activities. Therefore we could guess that there were the relationships between hemolysins and proteases produced by pathogenic vibrios.

    To determine the effect of body size on the clearance rate and ingestion rate of small intertidal bivalves, Glauconome chinensis, feeding experiments were conducted on individuals of 12 different size classes, from 4 to 16 mm in shell length. Relationships between morphological parameters were also determined. The clearance and ingestion rates of G. chinensis responded similarly to their body size, ranging from 1.3 to 28.2 mL/hr/ind. and from 24.0 to 458.5, ${\mu}gC/hr/ind$ ., respectively. Both rates increased significantly (p $(CR_w)$ and ingestion rate $(IR_w)$ decreased with increasing body size, with values from 1.0 to 3.1 L/hr/g and from 17.9 to 51.3 mgC/hr/g, respectively. The $CR_w$ of G. chinensis was intermediate compared to those of larger bivalve species. The clearance rate (CR) relative to flesh dry weight (FDW) of G. chinensis were fitted well to the power function: $CR=0.43\times(FDW)^{0.71}\;(r^2=0.89)$ . The exponent of fitting equation (0.71) of G. chinensis was higher than those of Mytilus edulis (Walne, 1972), Crassostrea gigas (Walne, 1972), and Placopecten magellanicus (MacDonald and Thompson, 1986).

    Pacific oysters Crassostrea gigas (Thunberg) were collected on April, 1999 and March­September, 2000 from Goseung Bay along the southern coast of Korea. The oysters tested cp;;ected from a depth of 0.5-2 m in which they cultured by a long line hanging method. The oxygen consumption rates (OCR) of oysters held under constant temperature and darkness (CC), were determined using an automatic intermittent-flow-respirometer (AIFR). Depending on holding periods after oyster collection, the experiments were divided into two groups: Group 7-d (held to ambient temperature for ca. 7 days) and Group 2l-d (held to ambient temperature for ca. 21 days). The OCR for Group 7-d single oyster displayed two peaks every day under CC, while Group 2l-d single oyster showed one peak every day. It is likely that the rhythmic patterns 02.6-12.8 hours) of the OCR in the Group 7-d single oyster may have been influenced by tidal currents at the sampling site. The rhythmic patterns (24.3-24.7 hours) in the Group 2l-d single oyster may have been shifted from two peaks to one peak each day under CC. The present study concludes that the OCR rhythm of wild oysters in nature is governed by two lunar-day clocks (24.8 hours); one driving one peak and the other driving the second peak. When oysters are subjected to the long-term CC conditions, one of the two-clock systems is depressed or only intermittently becomes active. Jpwever. the OCR rhythms by two to three oysters occurred arrhythmic patterns during the experiments and exhibited some evidence of weak rhythmicity of compared to those of a single oyster. It could be partly due to differences group effects.

    Temperature-dependent pharmacokinetics of ciprofloxacin (CIP) was studied in the cultured olive flounders, Paralichthys olivaceus, and black rockfish, Sebastes schlegeli using high performance liquid chromatography (HPLC) originally developed for quinolone determination from livestock. Pharmacokinetics of CIP was apparently affected by ambient water temperature. In a two-compartment model for flounders after oral dosage of 20 mg/kg, $K_{01},\;at\;13^{\circ}C$ and $23^{\circ}C$ were 4.18 and 1.20/hr, respectively. The $K_{10},\;T_{max}\;and\;C_{max}\;at\;13^{\circ}C$ were 5.574/hr, l4.37 ${\mu}g/mL\;and\;3.15{\mu}g/mL,$ respectively. The corresponding values at $23^{\circ}C$ were l2.84/hr, 15.39 ${\mu}g/mL\;and\;6.38{\mu}g/mL$ , respectively. The AUC, $T_{1/2} (\alpha)\;and\;T_{1/2}\;(\beta)$ were 278.23 ${\mu}g \cdot hr/mL$ , 0.24hr and 47.02hr at $13^{\circ}C$ and 3l7.8l ${\mu}g \cdot hr/mL$ , 0.30 hrs and 60.78hrs at $23^{\circ}C$ for the flounder, respectively. Similar CIP pharmacokinetics were revealed in the black rockfish after oral dosage of 20 mg/kg under the two water temperature regimes. These pharmacokinetical results have some implication in the optimal usage of recently introduced antibacterials in the farmed fish, which were primarily adapted for poultry and mammalian species.

    The aim of this study was to obtain the basic data on the fish school behavior change to approaching vessel and fish species identification by means of their swimming speed. The surveys were carried out for the juvenile southern blue fin tuna and other fish schools off Esperance, western Australia from January to March 1999. We observed changes of fish school behavior in response to the approaching vessel using 360-degree scanning sonar. The results showed that, a horizontal direction index used to quantify a change of fish school behavior did not identify dependence of a radial distance and a swimming speed. A Mann­Whitney test conducted using the horizontal swimming speed of both species identified by sonar specialists, did not reveal a significant difference.
