    DNA vaccine approaches have been applied to generate the protective immunity against various pathogens. However, the strength of immune responses induced by DNA vaccine is weak compared with conventional vaccines. The prime-boost vaccination using DNA vaccine and other viral vector has been suggested as one way to circumvent this limitation. In the present study, we used in vivo CTL activity assay to determine $CD^{8+}$ T cell-mediated immunity induced by prime-boost vaccination with a DNA vaccine ( $gB_{498-505}$ DNA) and recombinant vaccinia virus ( $VVg_{B498-505}$ ) expressing $gB_{498-505}$ epitope peptide (SSIEFARL) of herpes simplex virus type 1 (HSV-1) glycoprotein B (gB). The most potent in vivo CTL activity was induced in mice received $VVgB_{498-505}$ when both $gB_{498-505}\;and\;VVgB_{498-505}$ were used at priming step and boosted with the alternative vaccine vector expressing whole antigen protein (gBw). Priming with vaccine vector expressing gBw followed by the use of $VVgB_{498-505}$ at boosting step also induced strong in vivo CTL activity. We also examined in vivo CTL activity after immunization of mice with epitope-expressing vaccine vector at both priming and boosting step. Curiously, in vivo CTL activity mediated by $CD^{8+}$ T cells was strongly elicited at memory stage when animals were primed with $VVgB_{498-505}$ and subsequently boosted with $gB_{498-505}$ DNA. Because the use of $VVgB_{498-505}$ at priming followed by boosting with $gB_{498-505}$ DNA induced most optimal immunity, these results suggest that the order of vaccine type should be carefully considered when used vaccine type expressing only epitope for prime-boost vaccination.

    In this study, we investigated the role of toll-like receptor (TLR) and mitogen-activated protein kinase (MAPK) pathways involved in the tumor necrosis factor (TNF)- ${\alpha}$ and interleukin (IL)-6 expression after stimulation with purified protein derivatives (PPD) or native 38-kDa protein antigen (Ag) of Mycobacterium tuberculosis H37Rv in human primary monocytes. Both PPD and 38-kDa Ag significantly induced TNF- ${\alpha}$ and IL-6 in human primary monocytes. MAPK [extracellular signal-regulated kinase (ERK) 1/2 and p38] are rapidly phosphorylated in human monocytes stimulated with the PPD or 38-kDa Ag. Both p38 and ERK 1/2 activation are essential for PPD- or 38-kDa-induced TNF- ${\alpha}$ and IL-6 production. The inhibition of TLR2 and TLR4 by specific antibodies significantly abrogated the 38-kDa-induced secretion of TNF- ${\alpha}$ and IL-6, whereas blockade of TLR2, but not TLR4, was responsible for the PPD-induced TNF- ${\alpha}$ and IL-6 production in human monocytes. Collectively, these data suggest that the PPD and 38-kDa Ag differentially interact with TLR2 and TLR4, which in turn mediate an essential role for the early inflammatory immune responses during human tuberculosis.

    초록

    The envelope (E) glycoprotein of JEV is the major antigen to elicit neutralizing antibody (NAb) against JEV infection. In order to develop a rapid and safe neutralization assay system for evaluation of the JEV vaccine strains, we constructed JEV-pseudotyped viruses with JEV env genes (Nakayama-NIH, Beijing-1). The titers of JEV-pseudotyped viruses with NK and BJ strains were $4.0{\times}10^4IFU/ml\;and\;1.3{\times}10^5IFU/ml$ in Vero cell cultures, respectively. We have analyzed the neutralization activity of immunized mouse sera with JEV-NK and JEV-BJ pseudotyped viruses. The neutralizing antibody titers of NK and BJ (50% reduction of virus) were about 1:10,000 at each immunized sera. Compared with conventional plaque reduction neutralization test (PRNT), the method using JEV-pseudotyped virus has desirable advantages such as more rapid, easier, and non-biohazardous. This neutralization assay system might be useful to evaluate NAb activity against JEV vaccine strains or vaccine candidates.

    Flt3 Ligand (FL) and IL-6 are multifunctional cytokines implicated in normal hematopoiesis and ex vivo expansion of hematopoietic stem cells. Retroviral vectors are useful for stable expression of genes in many cells. Here, we aimed to produce retroviral vectors directing expression of human FL and IL-6 genes. Recombinant retroviral vectors containing human genes for FL and IL-6 were constructed using a retroviral vector pLXSN. Recombinant retroviruses were produced from GP2-293 cells with the aid of pseudo-envelope protein gene VSV-G, and efficiently transduced to a mouse stromal cell line OP9. Genetically modified OP9 cells clearly showed expression of human FL or IL-6 gene at the mRNA level determined by RT-PCR. Based on the results from ELISA, human FL and IL-6 were detected in the cell culture medium of OP9/FL and OP9/IL-6 cells, respectively. As the recombinant human FL and IL-6 proteins are successfully produced and secreted to the culture medium, this system can be useful in future application such as ex vivo expansion of hematopoietic stem cells and differentiation of embryonic stem cells.

    This study has focused on genetical patterns of rotavirus positives on diarrhea patients in Incheon. Fecal specimens collected from January 2002 to December 2004 from patients with diarrhea in seven localities in Incheon were screened for group A rotavirus by RT-PCR. To survey the prevalence of group A rotavirus infection and distribution of P (VP4) and G (VP7) genotypes of rotaviruses in Incheon, we performed the nested RT-PCR using rotavirus ELISA positive samples. Among a total of 4,865 samples investigated, 1,100 samples (22.6%) were positive. The predominant genotypes of 293 RT-PCR positive samples were confirmed as G4P[6] (101 samples, 34.5%) followed by G2P[4] (73 samples, 24.9%), G1P[8] (38 samples, 13.0%), G3P[8] (28 samples, 9.6%), G3P[10] (14 samples, 4.8%), G4P[8] (6 samples, 2.0%) and G9P[8] (6 samples, 2.0%). Mixed types and untypable types were 3.8% and 5.1%, respectively. The most prevalent types in 2002, 2003 and 2004 were G4P[6] (71 samples, 40.3%), G4P[6] (22 samples, 40.7%) and G2P[4] (15 samples, 22.2%) respectively. This is the first nationwide genoepidemiological study for determining the prevailing of genotypes of rotaviruses in Incheon. The genetic analysis for investigating sequence variations among group A rotavirus strains in this study would provide the useful information to establish the preventing strategy of human rotaviruses and of developing vaccines which will be used in Korea.

    This study has focused on genetical patterns of rotavirus positives on diarrhea patients in Incheon. Fecal specimens collected from January 2002 to December 2004 from patients with diarrhea in seven localities in Incheon were screened for group A rotavirus by RT-PCR. To survey the prevalence of group A rotavirus infection and distribution of P (VP4) and G (VP7) genotypes of rotaviruses in Incheon, we performed the nested RT-PCR using rotavirus ELISA positive samples. Among a total of 4,865 samples investigated, 1,100 samples (22.6%) were positive. The predominant genotypes of 293 RT-PCR positive samples were confirmed as G4P[6] (101 samples, 34.5%) followed by G2P[4] (73 samples, 24.9%), G1P[8] (38 samples, 13.0%), G3P[8] (28 samples, 9.6%), G3P[10] (14 samples, 4.8%), G4P[8] (6 samples, 2.0%) and G9P[8] (6 samples, 2.0%). Mixed types and untypable types were 3.8% and 5.1%, respectively.
