    초록

    In order to investigate biochemical activities of harvested tobacco leaves, photosynthetic rate, soluble protein contents, and peroxidale activities were analysed during different maturing period. Physiological activities of harvested leaves during maturing period were higher in topped than those of non-topped plants. Chlorophyll content and photosynthetic rate in both topped and non-topped plants decreased at 4 days and 3 days before harvest, respectively. The chloroplast numbers in topped and non-topped plants decreased at 3 days and 5 days before harvest, respectively. Changes of soluble protein and total RNA contents showed similar patterns during maturing period. Soluble protein contents were slightly decreased from 5 days before harvest in topped plants, but decreased drastically from 3 days before harvest in non-topped plants. Not much changes were found in total RNA contents in topped plants until 2 days before harvest, and it was largely decreased after 5 days before harvest in non-topped plants. The peroxidase activities drastically decreased in topped plants and increased in non-topped plants after 3 days before harvest during maturing period. The largest change of biochemical activities in tobacco leaves during maturing periods were observed at 3 days before harvest.

    초록

    The vein-necrosis strain or potato virus Y (PVY-Vff) and black shank (Phytophlhora parasitica roar. nicotianae) causes severe damage on burley tobacco(Wicotiana tabacum L.) in Korea, A new burley tobacco resistance to PVY and black shank, KB 110, was developed by Korea Ginseng and Tobacco Research Institute. It was developed from the cross of Burley 21 with TC 591 in 1990, and was backrossed to Burley 21 in the following season. TC 591 has resistance to PVY and moderate resistance to race 0 of black shank, but it is susceptible to tobacco mosaic vim (TMV). KB 110 was evaluated for its resistance to PVY, TMV and black shank in the greenhouse and at fields for preliminary and performance trials. KB 110 which has secreting glandular trichomes was resistant to PVY-VN, TW and black shank. It had an erect growth habit and two more leaves per plant than that of Burley 21, and matures two to three days later. It yielded approximately 3 percent more cured leaf than the standard cultivar Burley 21, but other plant characteristics were very similar to those of Burley 21. It had acceptable standards for chemical and physical characteristics of lured leaf on regional farm test in 1995-1997. KB 110 produced average yields of good quality tobaccos and was appeared to be resistant to PVY inwhere occurrence of the virus are severe chronic at burley growing area.

    초록

    This study was conducted to investigate the effect of fly ash on the yield and quality and to determine the optimum application amount of fly ash for tobacco(Nicotiana tabacum L). Two kinds of fly ash, anthracite and bituminous coal, were treated with different levels of 0, 20, 40, 60 MT/ha. Dry weights of tobacco at middle and topping growth stage were increased with application of fly ash, showing the highest dry weight at 40 MT/ha in both kinds of fly ash. It was showed that the bituminous coal had a little more effective for yield than that of anthracite. Comparing with the control, yields of tobacco applied with fly ash were significantly increased about 17.7% and 17.1% by the application of bituminous coal and anthracite, respectively. Quality of flue-cured leaves was better by application of fly ash than that of the control. The quality index was given the highest at 40 MT/ha for bituminous coal increasing by 24.6% and at 60 MT/ha fur anthracite increasing by 13.4% compared with the control. The economical efficiency considered of the yield and quality of tobacco was the highest at 40 MT/ha of bituminous. Soil pH, contents of available P2O5, organic matter, exchangeable Ca2+ and Mg2+ of soil during the growing season were increased by application of fly ash, showing more effectiveness in bituminous than that in anthracite. By the application of fly ash, the nutrients availability and the acidity of soil were reformed and they caused significantly the increase of growths yield, and quality of tobacco. By the application of lime reforming soil acidity, growth response, yields and quality of tobacco were not increased compared to the control, although the effect of reforming soil pH was remarkable.

    초록

    The inluence of styrofoam-panel thickness of floating frame on the seedling growth of flue-cured tobacco(Nicotiana tabacum L.) in the newly developed direct-seed float system was investigated . Floating frame used instead of styrofoam-tray consisted of styrofoam-panel (106×108cm) for floating, eight holes(1.Sx2S.0 cm) to uniform the water, and nutrition supply far seedling and water and nutrition absorption cloth placed on the styrofoam-panel. Each floating-frame may lay eight plastic-trays on it. Eight grades of styrofoam-panel thickness from 26 mm to 46mm and two kinds of media were used in the greenhouse system. Dry cells were found from the styrofoam-panel thickness of 43mm in carbonized chaff, compost and original soil rate of L3-3(v/v) media, and of 37mm in peat and perlite rate of 8-2(v/v) media. The thinner styrofoam-panel produced more tender and succulent seedling with the more trunk and the less root weight. When considering the appearance of dry cell, seedling growth, producing healthy tobacco seedlings, and utilization of styrofoam-panel for two kinds of medias the ideal styrofoam-panel thickness were suggested to be around 34mm for flue-cured tobacco reeling production in the newly developed direct-seeding float system.     초록

    Karyotype of apterous green peach aphid (Myzus persicae SuIBer) populations occurring in tobacco fields and their resistance to insecticides were investigated. All of the 16 clones collected from tobacco fields had same chromosome number of 2n=12. Among them, 5 clones had the normal karyotype, regardless of color morph, but chromosomal translnations between #1 and #3 autosomal chromosomes were noted in the other 11 clones. All of the aphid clones were highly resistant tolambda cyhalothrin, a pyrethroid pesticide, having over 20 times higher concentrations of LD90 than recommended concentration (10ppm) for the aphid. However, their resistance to acephate, demeton 5-methyl and pirimicarh varied depending on the clones, among which green-colored clones were generally more resistant to the pesticides than red-colored ones. No association was found between the insecticide resistance and the karyotype of the aphid in this experiment.

    초록

    Viral coat protein (CP) encoding cDNA with artificial start and stop codons was synthesized by reverse-transcriptase polymerase chain reaction (RT-PCR) from the Korean isolate of potato virus Y-vein nectrosis strain (pVY-VN). To make PVY CP cDNA to untranslatable form, three stop codons were inserted near the start codon by "megaprimer-PCR" method. The untranslatable CP cDNA was subcloned to plant expression vector and transferred to N. tabacum cv. NC82 by Agrobacterium-mediated transformation. Highly resistant plants to PVY infection were screened, based on symptom development after mechanical virus inoculation. By genomic PCR and Southern blot analysis, one or more copies of the untranslatable CP gene were found in all transformants. From northern blot analysis, highly resistant transgenic lines had very low level of CP transcript but susceptible lines had high level, suggesting resistance to PVY infection should be related to RNA-mediated mechanism.mechanism.

    Semivolatile components in mainstream smoke of cigarette attached the different type filter were analysed. Based on the main peak of GC profile, benzene, toluene, acetic acid, limonene, acetamide, pyridine, nicotine, phenol, pyrrole, and furfuryl alcohol were identified. The amount delivered of semivolatile components by each filter was evaluated with the ratio of areas of sample vs area% of ISTD. By comparing dual and triple filter to mono filter, the delivery amounts were 52% benzene, 62% toluene and 74% benzene, 84% toluene, respectively. The delivery amount of limonene which was known of tobacco taste component, were 52%, and 93% by dual and triple filter, respectively. Also, delivery amounts of acetic acid which was one of acidic compounds in smoke, were 86% and 83% by dual and triple filter, respectively. When adding the same amount of active carbon, the amounts delivered of vapour phase such as benzene and toluene by dual filter were lower than that of triple filter. But the delivery amounts of acetic acid and phenol by dual filter were higher than that of triple filter. This results assume that the pH of active carbon in filter affect to the adsorption or absorption of triacetin during filter making process.

    초록

    This study was carried out to investigate the influence of triacetin(TA) on some phisico-chemical properties of cigarette filter rods. To find the effect of TA on physical properties, we made six kinds of filters varying the TA levels(0%, 2%, 4%, 6%, 8%, 10%) and measured the hardness, resilience and roundness. The hardness of filter rods was increased from 83.2% to 94.6% with increasing the TA level hut the roundness was not affected by TA levels. After manufacturing the cigarette with the filters, we have analysed the delivery amount of phenol and TA with the content of TA. The amount of phenol was decreased from 20.6 $\mu\textrm{g}$ /cig. to 12.0 $\mu\textrm{g}$ /cig. with increasing the TA level of the filters. The decreasing of phenol amount was thought to be caused by solution effect. Also the TA delivery of TPM was increased from 0% to 5% of TPM with increasing the amount of TA. We have found the interesting phenomena that ventilation rate was also decreasing up to 6oye with increasing TA. It seemed to be due to the encapsulated pressure drop and the reduction of the plugwrap porosity which also caused by the smear with TA on the surface of plugwrap. From this results, we could find that not only the hardness, resilience and ventilation rate but also the contents of phenol and TA in cigarette smoke was affected by the TA levels on the cigarette filter rods.

    Cigarette smoking is known to suppress both 1-methy14-phenyl-155,Ltetrahydropy-ridine (MPTP)-induced parkinsonism and idiopathic Parkinson's disease (PD). However, the precise mechanism underlying its protective action against PD is not clearly elucidated yet. In order to find possible clue on the mechanism of protective action of smoking, we investigated the inhibitory effect of cigarette smoke components on rat brain mitochondria1 monoamine oxidase B (MAO-B), responsible enzyme for the activation of MPTP to its toxic metabolitesr and identified the components having an inhibitory potency on this enzyme from cigarette smoke. Total 31 eligible constituents including nicotine were selected from cigarette smoke condensates via solvents partitioning and silica gel chromatographic separation, and inhibitory potencies of 19 components on MAO-B were determined. Hydroquinone and methylcatechol, the phenolic components, showed the strongest inhibitory potencies on MAO-B activity in the components tested. 3,4-Dihydroxybenzylamino, myosmine and indole in basic fracton, eugenol in phenolic fraction, and farnesol in neutral fraction also inhibited the enzyme activity dose-dependently. Among tobacco alkaloids tested only myosmine was effective for the inhibition of this enzyme. These results suggest that the decrease in MAO-B activity by such components derived from cigarette smoke seems to be related to the suppression of MPTP-induced neurotoxicity and to the less incidence of Parkinson's disease in smokers than in nonsmokers.

    Nonvolatile polyhydroxyalkylpyrazines(PHAPs) are known to degrade volatile pyrazine compounds having positive aroma and sensory attributes during cigarette smoking. In this paper, the content of PHAPs in burley tobacco leaves toasted to different levels as temperature and time study were determined by high performance liquid chromatography, and was also investigated effects of added sugars, sucrose, glucose or fructose, on the generation of PHAPs during toasting. The addition of glucose or fructose as casing sauce in burley tobacco leaves resulted in significant increases, especially 2,6-deoxyfructosaBine from glucose added leaves and 2,5-deoxyfructosazint from fructose added leaves, in generation of PHAPs during toasting, while control and sucrose added leaves did not observed in generation of PHAPS. Formation rates of PHAPs in glucose or fructose added tobacco leaves, but not sucrose, showed a strong dependence on both toasting temperature and time.
